Computational Assessment of Modified Antifreeze Glycoproteins on Ice Nucleation

چکیده

Antifreeze glycoproteins (AFGPs) found in various fish are used by the organisms to prevent freezing. While these compounds have been studied for their ability bind to, and complete crystallization of water, exact mechanisms which AFGPs freezing still undetermined. Therefore, building upon our previous work, this study uses molecular dynamics simulations assess effects hydroxyl group separation distance on AFGP ice nucleation activity. Water droplet showed that modified structures containing distances smaller than ~3.0 Å lost crystallization. Furthermore, 7.327 6.160 was correlated with a promotion nucleation, as demonstrated changes energy system. This supports notion distance, therefore, geometry characteristics between groups located saccharide play key role inhibition properties compounds.

Janus effect of antifreeze proteins on ice nucleation.

The mechanism of ice nucleation at the molecular level remains largely unknown. Nature endows antifreeze proteins (AFPs) with the unique capability of controlling ice formation. However, the effect of AFPs on ice nucleation has been under debate. Here we report the observation of both depression and promotion effects of AFPs on ice nucleation via selectively binding the ice-binding face (IBF) a...

Antifreeze glycoproteins from Arctic fish.

Four species of fish in the Barents Sea of the Arctic, north of Norway and Russia, were caught and examined for the presence of antifreeze proteins in their blood sera. Only one species of Arctic fish, the polar cod (Boreogadus saida), was found to contain an antifreeze protein. Antifreeze glycoproteins were isolated and characterized from the blood serum of the Arctic fish, and their structure...

Production of Ice Nucleation Deficient (Ice-) Mutants of the Epiphytic Strains of Erwinia herbicola

To mutate the Ice Nucleation Active (INA) gene in Erwinia herbicola strains, Tn-5 transposon carried by Psup2021 plasmid was used. This plasmid was transferred to the bacterial cells by electroporation. Electrotransformation was carried out for 2.5 ms at 1800 v and 1 mm distance between the electrodes. Polymerase chain reaction was used for determination of presence or loss of INA gene, using a...

Role of ice nucleation and antifreeze activities in pathogenesis and growth of snow molds.

ABSTRACT We examined the ability of snow molds to grow at temperatures from -5 to 30 degrees C and to influence the growth of ice through assays for ice nucleation and antifreeze activities. Isolates of Coprinus psychromorbidus (low temperature basidiomycete variant), Microdochium nivale, Typhula phacorrhiza, T. ishikariensis, T. incarnata, and T. canadensis all grew at -5 degrees C, whereas Sc...

Direct Observation of Postadsorption Aggregation of Antifreeze Glycoproteins on Silicates

The adsorption of antifreeze glycoproteins (AFGP) from aqueous solution onto two different silicate minerals, muscovite mica and amorphous silica-titania, has been directly observed for the first time in situ using atomic force microscopy (AFM) and optical waveguide lightmode spectroscopy (OWLS). The former yields the lateral distribution and heights of adsorbed single molecules and massed adso...
